As in bacteria that convert nitrate to nitrogen? You can do it, but you would need media that is extremely porous to house the anaerobic bacteria and a very low flow rate at or below 50 gph. You need to create an environment in the canister with as little oxygen as possible.

Possibly, but you would need a valve to cut back the flow rate. The whole system and the piping would have to be designed to reduce oxygen and flow as little water as possible. Its not easy to accomplish.

Not really as shown above. Something like a large surface area mattenfilter dividing a whole sump tank is the most easy way to ensure this is actually happening. Plants and algae scrubbers probably do as well or better though.
